It was founded in 1836 by people who came from Ste-Anne-de-la-Pérade following to the east along the Sainte-Anne.

The name of Saint-Casimir recalls the memory of the notary Joseph-Casimir Dury (sometimes, one writes Duret or even Déry) of Sainte-Anne-de-la-Pérade, one of the benefactors of the parish who financed the construction of the parish church. This imposing Gothic-style church was decorated by Raphaël Giroux, a pupil of Thomas Baillargé.

In turn, the church, and the town, is named after Saint Casimir, a patron saint of Poland, Lithuania, and youth.

To the east of the village, there is a fascinating cave called Devil's Hole. The cave, eight thousand years old and 980 meters (3220 feet) long, is the second longest known cavity in Quebec.

At the beginning of New France, the territory of Saint-Casimir was part of the seigniory of Grondines, one of the oldest in Quebec, founded in 1637, and of the parish of Saint-Charles-des-Grondines.

The seigniory was granted by the Compagnie de la Nouvelle-France to Madame de Cambalot, Duchess of Aiguillon.
In 1646, the governor of New France, Monsieur de Montmagny, granted the nuns Hospitallers possession of the territory of Grondines, but in 1683, the Hospitallers sold the seigniory to sieur Jacques Aubert. The seigniory then changed hands several times, but it was around 1830 that Mr. Peter Burnet, a lumber merchant from Quebec, established a saw and flour mill there, at the confluence of the Noire and Sainte-Anne rivers (today today, it's the junction of Tessier and Chemin de l'Île Grandbois streets).

During the 1760 census, there were already about thirty houses in the area, dotted along the banks of the Sainte-Anne River, but it was from the construction of the mill that the municipality developed.

Towards the end of 1835, the residents addressed a request to the Archbishop of Quebec, Monsignor Signay, asking him to erect a parish. The response was positive, and the new parish was canonically constituted in 1836.

On March 22, 1997, a collective suicide took place in Saint-Casimir, when five followers of the Order of the Temple of the Sun, three of whom were of French origin, set fire to the house, where they were and are died in the flames.
They are Didier Quèze, 39 years old; Chantal Goupillot, 39; Suzanne Druau, 63; Bruno Klaus, 49; Pauline Rioux, 54 years old.
